Hybrids and Hybridization Oh, Sang-Hun [1], Suh, Hwa-Jung [2], Youm, Jung-Won [3], Park, Chong-Wook [4]. Origin of Calanthe x kibanakirishima (Orchidaceae). Calanthe is a large genus in Orchidaceae with about 200 species broadly distributed in tropical and subtropical areas of Asia, Africa, Australia, and Central and South Americas. Species of Calanthe are widely cultivated as an ornamental because of showy flowers. In Korea, five species have been known in the southern coastal regions, Ulleung Island, and Jeju Island, but the populations are under threat of extinction due to overexploitation and habitat destruction. Recently distribution of C. x kibanakirishima was newly reported in Korea. This rare species has been known to be a putative natural hybrid between C. sieboldii and C. aristulifera based on morphology. To test the hypothesis of hybrid origin of C. x kibanakirishima, we located a population on a remote island in Sinan-gun, southwest of the Korean Peninsula, where C. x kibanakirishima grows together with C. sieboldii and C. aristulifera. DNA sequences of nuclear ITS and three chloroplast regions (atpF-atpH, psbK-psbI, and trnH-psbA) were examined from three individuals of C. x kibanakirishima, four individuals of C. aristulifera, and 30 individuals of C. sieboldii. Samples of C. discolor, C. bicolor, and C. reflexa were also included. Sequence analysis of the combined chloroplast DNA data results in ten haplotypes from the six species. All three individuals of C. x kibanakirishima have an identical cpDNA haplotype, and it is unique, not shared with any species. Results of this study suggest that neither C. sieboldii nor C. aristulifera is a maternal progenitor of C. x kibanakirishima. Phylogenetic analysis of ITS sequences suggests that C. aristulifera is nested within C. x kibanakirishima. Four polymorphic sites were found in C. x kibanakirishima. However, we have found no evidence that the ITS sequences of C. x kibanakirishima are homeologues derived from C. sieboldii and C. aristulifera.
