Pteridological Section/AFS Patel , Nikisha Rashmi [1], Barrington, David [2]. Origins of the unknown North American tetraploid Phegopteris (Thelypteridaceae). The small fern genus Phegopteris (Thelypteridaceae) comprises three to five species, plus an undescribed apogamous tetraploid whose origins are unknown. Previous work on populations of the unknown tetraploid from Quebec and Vermont have yielded two hypotheses for its origin. On the basis of morphological intermediacy, the species in question was proposed to have originated from hybridization between Phegopteris hexagonoptera (a sexual diploid) and Phegopteris connectilis (an apogamous triploid), but isozyme analysis and review of morphology has suggested that the unknown tetraploid does not include a genome of the former. The present study sampled Phegopteris more widely, including a previously unsampled east-Asian diploid cytotype of P. connectilis . Sequences retrieved from cloning of ITS indicate that the unknown tetraploid contains at least two genomes. Both chloroplast (trnL-F, RPS4, rbcL, and psbA) and nuclear (PGI and ITS) markers reveal that the proposed diploid progenitor P. hexagonoptera is not one of these genomes.
