Systematics Section/ASPT WILSON , CAROL A [1]. Clade recovery in the morphological-divergent onco irises using low-copy nuclear and cpDNA sequence data. Iris sect. Oncocyclus is a group of about 31 bearded species that are distributed in the Caucasus south to Iran and southwest into Turkey, Syria, and regions adjoining the eastern shores of the Mediterranean Sea. Plastid genome and RNAseq data was used to develop primers for Sanger sequencing of cpDNA and low-copy nuclear regions with promising levels of variation. This study included 33 taxa representing 26 species and utilized more than 8,000 bp of cpDNA and about 2,000 bp of low-copy nuclear data to investigate relationships among the Oncocyclus species. Sequence divergence of the plastid genome is low, nuclear divergence is moderate, and morphological divergence is high. Plastid data reveal biogeographic patterns with clades recovered that mostly represent Caucasus species, species from eastern Turkey and western Iran, or species from Syria and southern Turkey. Some of these topologies were shared with nuclear trees although there is also some discordance between marker types. There is greater variation in nuclear compared to plastid data but the level of branch support across trees is similar. Even though relationships are not completely resolved, several clades are consistently recovered that begin to elucidate the evolutionary history of this group.
