Systematics Section/ASPT Hoose, Dalton [1], Willyard, Ann [1]. Designing an array to target orthologous low copy nuclear genes for Next-Gen Sequencing in Pinus. The recently published assembly of the Pinus taeda genome provides an improved platform for selecting orthologous low copy nuclear genes that are informative for intra- and interspecific comparisons. In addition, a recently published dense genetic linkage map for P. taeda provides an important filter because genes that map have a higher probability of being low copy. We used these data, along with earlier genes identified from expressed sequence tags, to create an array that should be useful for all of the hard pines (subgenus Pinus). The selected genes will be used to design biotinylated probes to target specific sequences in order to create a species tree for the P. ponderosa species complex.
